摘要

通过对天然多糖进行化学修饰

制备具有ATRP引发位点的多糖引发剂

然后通过原子转移自由基聚合

制备以天然多糖为骨架

以不同链长的阳离子聚合物为侧链的阳离子非病毒基因载体。对合成的材料进行细胞转染、细胞毒性测试

并对其它各种性能进行表征。各项表征的结果证明基于ATRP法合成的多糖基因载体能够很好地络合DNA

并且具有良好的PH缓冲能力以及生物相容性

与PEI和商业化转染试剂相比

转染效率更高

细胞毒性更低

在基因治疗中具有很好的应用前景。

Abstract

Through chemical modification of polysaccharides

we developed bromoisobutyryl- terminated polysaccharide initiators for subsequent atom transfer radical polymerization. A series of new cationic polymers composed of biocompatible polysaccharide backbones and polycation side chains of different length were designed. Cell transfection

cell toxicity

and the other properties of the material were characterized. The results of the characterization indicated the cationic polymers can condense DNA very well and have good buffer capacity and biocompability. The high gene transfection efficiency and cell viability of polysaccharide-based polycations compared with PEI and commercial transfection reagent demonstrate it has great potentials as efficient vectors in future gene therapy.
